Supported by key projects of the National Natural Science Foundation of China, the Institute of Biology and Chemical Engineering at Tsinghua University has made a breakthrough in biotechnology by successfully developing China's first intelligent, multifunctional enzyme—MBP (maltose-binding protein) fusion heparinase. Following six months of pilot testing, the research team has consistently produced ultra-low molecular weight heparin with an average molecular weight ranging from 2000 to 3000, reaching international advanced standards.
Enzymatic catalysis is a cornerstone of green chemistry, and for many years, China has heavily relied on imported low molecular weight heparin, a critical drug for preventing and treating thrombosis. This dependency has led to high costs and limited domestic production capabilities. The development of this new enzyme represents a significant step forward in both scientific and social terms.
The research team utilized advanced biochemical technology to create a novel process for producing highly efficient, intelligent, and multifunctional MBP fusion heparinase. They introduced a fluorescent protein signal to the enzyme, enabling real-time tracking. Additionally, they incorporated an easily separable MBP protein, allowing the heparinase to be purified in a single step, significantly improving efficiency and purity.
After half a year of industrial trials, the MBP fusion heparinase demonstrated excellent performance, effectively degrading heparin and producing ultra-low molecular weight heparin with precise control over reaction time and enzyme dosage. Notably, the enzymatic process is gentle, offering advantages such as high yield, environmental friendliness, and the potential for continuous production. The product and the enzyme can be easily separated, allowing the enzyme to be reused, which reduces both enzyme cost and overall production expenses.
This innovative enzyme has already secured three patents and was awarded the Silver Prize at the 17th National Invention Exhibition in November 2007. Looking ahead, the research group is now applying this intelligent, multifunctional enzyme to the development of phosphotransferases and industrial enzymes. In particular, when used in the fine chemical industry for food additives, the biological method enabled by this enzyme offers a safer alternative to traditional chemical processes, which are often toxic. Unlike the monopolized methods from Japan and South Korea, this approach allows real-time monitoring of enzyme activity, ensuring a clean, mild, and pollution-free reaction process.
